Association of Quantitative Characteristics with Growth Hormone Gene (GH Gene) in Kerinci Duck Using PCR-RFLP Method

This study aims to determine the association of quantitative characteristics with growth hormone gene (GH gene) in kerinci duck using PCR-RFLP method. Samples total used was 96 Kerinci ducks consisting 43 males and 53 females blood samples. DNA extracted protocol Genomic Purification Kit from Promega then amplified by PCR a pair primers5'-CAA GGA ACA GAG GGT TTC CA-3' Revers : 3'-GGG AGA TAG GGC AAA CAT CA-5', length product 855 bp. The amplification cut restriction enzyme AluI AG↓CT cutting site. Growth hormone/AluI fragments were electrophoresed 1.5% agarose gel visualized doc gel. Data collected includes body weight, weight gain, measurements, ducks. differences as well between genotypes tested T-test. determinants size shape analyzed PCA. showed that sizes male significantly different (P<0.05) than female GH|AluI is polymorphic three genotypes, i.e, +/+ (49%), +/- (39.6%), -/- (11.5%), two alleles, namely (+) 69% (–) 31%. Conclusion: higher female. identifier sternum, shank lengh, circumference, while wing length. GH Genes|AluI polymorphic. |AluI associated best genotype (+/+).
